Well, the fact he said no the last time, and ended up in doing it anyway, pretty much precludes any Guarantee that the next one will definitely be directed by someone else.
After all, Roger Moore made a career out of saying 'this will be my last', only to be enticed back into the fold. (Without any surprise, I might add)

Mendes does sound very positive, which, given some of the behind-the-scenes drama on Spectre, is encouraging.

I agree with Harmsway. You don't hear a hint of stress or fatigue in Mendes' voice, in fact just the opposite. I hear a tone of enthusiasm that does not sound manufactured. This is very encouraging.

Does he really rule it out?
I can't help but notice him use the word "probably" in the interview.
He was much firmer when he said he wouldn't be doing Bond 24, but then changed his mind.
Personally, I wouldn't rule him out, not based on this statement at this stage of making Spectre.

These productions are behemoths that eat up all your life. It´s perfectly understandable that he does not want to commit to the next one already. Also, it would be unwise for it would take him out of consideration for other projects in the next years.
In the end, if SPECTRE is received as well as SKYFALL (or better) he will return (with an ever bigger paycheck). If not he can always say: hey, I´ve done two back-to back, I want to do something else now.

Unfortunately I didn't catch up with the interview, but I get the gist of the Bond part of it from the posts above. Since then, I have come across a quote from Sam Mendes about SPECTRE found in, of all places, the online version of Ecumenical News. I don't know if this was part of the BBC interview, but here he says as follows;
"Spectre provides a kind of culmination to the three previous films while developing a backstory that's been largely unexplored until now. There is an opportunity to delve into how Bond came to be. What made him? And who were the people who affected him along the way? You're sort of telling the story backwards of how Bond became Bond."

Probably one of the broadest hints we've had yet that the new film ties up some loose ends from CR, QoS and SF, whilst delving into Bond's past, which we're already aware will happen from the trailers.
