There are so many to choose from, but I will have to go for the final scene, just because the hairs on the back of my neck stood on end, when DC delivered "that line".
And then the James Bond theme kicked in!!!
Great stuff.
How about you?

1. Bond shoots a guy in uber-cold blood style. Really gets down to what Bond's job really is.
2. "Yes... considerably", not so much a one liner as a retort, but Craig's delivery of that line had such bite to it. Perfect.
3. The Gunbarrel. The perfect summation of what I was thinking in theaters a second earlier. "Bond is back, bigtime!"

African Rundown - sheer cinematography heaven. Shower scene - David Arnold's "vesper" made hairs on the back of my neck stand up, the little dialogue and simplicity of the scene is what made it. The PTS was so well shot in black and white, sharp dialogue, brilliant Fight scene. Torture sequence and that final scene where you see his shoes step up on the steps and pan to DC ushering those final lines.

- The shower scene
- The poisoning (no pun intended but it was a heart stopper)
- The time on the beach and in the bedroom but before the finale

- the poker scene with Dimitrios in the Bahamas
- Bond and Vesper meeting for the first time on the train
- Le Chiffre torturing Bond aboard his barge
- anything with Mendel the Swiss banker!
But for me, the scene that excited me the most and really made the hairs stand up on the back of my neck was the final scene. All the excitement and tension building up to the "Bond, James Bond" line, and the theme music kicking in

Yes. Many to choose from.
Off the top of my head in no particular order:
- Embassy after the crane fight. That's "taking care of business".
- Confrontation with M.
- Winning of the DB5.
- The leverage move in the Body Exhibit.
- The train and, yes, Vesper was "the money". Best exchange ever.
- Stairwell. Surprisingly brutal.
- The shower. Tender and not in a seductive manner. He cared.
- Final trump at the big game.
- Dinner with Vesper.
- Dash through the square.
- Last shot. That's appealing not just for being Bond, but hasn't EVERYONE wanted to have that moment or similar over someone and truly put them under your thumb? The bully from grade school. The supervisor at work. The guy who stole your parking spot.
It was both payback and establishment of character and the perfect ending, if not a wrapping up to the film.

Other stand outs are the train ride with Vesper and the cab ride with Vesper.

- The PTS. I love Noir film and this is as close to Noir as James Bond can be
- The opening credit. I didn't like "You Know My Name" at first but after seeing it with the opening credit, it feels right. Daniel Craig looks unbelievably cool as he walks towards the screen, cards flying, and "the coldest blood runs through my veins, you know my name! (you know my name)...." plays in the background.
- The Africa chase scene. That jump from crane to crane was breathtaking. And for the first time ever I'm actually afraid for James Bond's life.
- The train scene. Classic. It reminds me of Cary Grant style films. The banter between DC and EG oozes a lot of chemistry.
- The stairwell scene. What a brutal fight which has me worried for JB and Vesper. I think this is one of the reasons why I like "Casino Royale" in that I know James is just a man and he can get hurt in his job if he's not careful and smart.
- The shower scene. Understated acting by both DC and EG.
- The poisoning scene. Again, I can't believe I actually thought JB is going to die. But that's credit to the writers and actors for being able to sell JB's vulnerability.
- The last scene. The brutal shooting of Mr.White feels satisfying after the terror he puts JB through. And that last line delivery by DC is just perfect.

I think that Bond is very similar in FYEO when persuading Melina to go back to Greece, while on the horse & carriage.

For right now, I'll have to go with the train scene, an instant classic.

Honorable mentions go to the train conversation with Vesper, "That last hand nearly killed me!", "That's because you know what I can do with my little finger!", the PTS, the free running chase, Bond taking Solange to "his place", the parking attendant bit, "Stephanie Broadchest"... oh hell, the whole film is fantastic!

But also, oddly enough, the quiet little silent scene where Bond's trying on his new dinner jacket and turns to see Vesper watching him. The range of emotions that flicker across his face!

1) The end of the Miami International sequence. When the bomber was gloating over Bond's arrest and about to trigger the detonator, I honestly thought, "Oh, no..." but then when he realises the detonator's on him and we cut back to Bond and him smirking as it goes off. Classic, and a prime example of what a swine Bond can be at times.
2) The end of an Aston Martin. Much as I love Astons and how much it pains me to see one get wrecked (I winced at the doc about the ice chase in Die Another Day when the stunt driver hit an iceberg and pretty much stuffed the nose of the Vanquish), the destruction of the DBS was brilliantly done. I remember the last time I went to see it, the guy sitting next to me whispered to his girlfriend, "Ouch!" as the car finished rolling. I don't know if he was referring to Bond or the Aston.

1.The Parkour Chase:Brilliantly done. One of the most exciting action scenes in Bond films & in film history which can't be topped anytime during the rest of the movie (thankfully the filmakers don't try)
2.Aston Martin DB5:Very well played out scene & when Daniel Craig throws the keys out of his hands, that's the moment I accepted him as Bond.
3.Miami Internation Airport:A very high-paced tense action sequence which ends perfectly with that little grin on Bond's face when the bad guy gets blown up.
4.Bond trying on his tuxedo:Perfectly iconic scene & DC just looks embodies the tuxedo.
5.Poisoning scene:Simply had me on the edge of my seat.
6.Torture scene:DC is simply amazing in this seat & it's quite scary, but also quite hilarious at the same time & I had 2 keep at least one hand over my "stuff" for the rest of the movie.
7.Final Scene:Only TSWLM's ending is in the same league as when DC says that iconic phrase so perfectly with one of the best versions of the Bond theme 2 accompany it.

2. The torture scene: While MIA is favorite and the one I was most looking forward to, this one caught me by suprise. I can't say I wasn't looking forward it, but I wasn't really dying to see it. I was blown away by Craig's performance and it did live up to the early hype from filmmakers that Craig was amazing in it.
3. The embassy shootout: I think it flowed better without the dialogue that was in the script. I love the breakneck pace. The energy in the sequence is insane and you can really feel the shock and suprise that the nambutu troops are in at finale.
4. The Poker games: I am a fan of Texas Hold 'em even before CR. I love the poker in the film, all the poker scenes. This is where Mads really shined as Le Chiffre. It was the subtle things he said and did. It was all in his eyes. Like when Bond leaves the table after ordering his martini you see as Le Chiffre follows him with eyes, all fumming like he's ready to explode. And when he's defeated by Bond he just stares at the cards on the table, he never looks at Bond. It's like he has been punched in the gut and you can see a small red tear starting to leak from his eye just before he stands up to leave.
5. The sinking house: It's a great sequence. The modle work is amazing and the fight with Gettler and his goons is classic. It's Bond his most brutal since LTK.

I am interested to know whether or not you knew that the bomb was on the bomber. The clue we get is that Bond grabs the back of his trousers while they are fighting on the truck.

You know, from what I remember of my first viewing of Casino Royale (I was getting better at spotting stuff on my subsequent viewings), I don't think I actually noticed Bond grabbing the back of the bomber's trousers.
It was like Richard Branson's cameo - I never noticed him at first. I think I was too caught up in the sequence. I did have an inkling that there'd be a twist (obviously, the bomber couldn't have succeeded, as otherwise it'd be a pretty short movie).
