Dryden: How did he die?
James Bond: Your contact? Not well.
Dryden: Made you feel it, did he? Well, you needn't worry. The second is... [easier]
[Bond shoots Dryden]
James Bond: Yes... considerably.

Fill in the blanks of Dryden's sentence and you'll get it:
Dryden: How did he die?
James Bond: Your contact? Not well.
Dryden: Made you feel it, did he? Well, you needn't worry. The second is... [easier]
[Bond shoots Dryden]
James Bond: Yes... considerably.
Beautiful line! Especially since Bond really means 'Yes this kill is considerably easier given the fact that I hate you.'

Just what the hell does Bond mean in the opening sequence when his target says "The second time is..." and Bond replies with "Considerably"?
Bond had just gotten laid and his performance was less than satisfactory. Dryden was telling Bond that the second time is much better.
Or did I misread the scene?

He says it as a statement of fact, rather than the gag it's supposed to be- he's answering Dryden saying that he doesn't know him very well: Bond knows where Dryden's gun is, which is a start towards knowing him.
I don't understand this comment. How is the line - "I know where you keep your gun." - supposed to be a gag, considering the noir setting of the scene?

I have the same question. He says it as a statement of fact, rather than the gag it's supposed to be- he's answering Dryden saying that he doesn't know him very well: Bond knows where Dryden's gun is, which is a start towards knowing him.

Dryden: How did he die?
Bond: Your contact? He died on the spot
....
Dryden: Well, you needn't worry. Your second kill is...
Bond: Yes. He died a much easier death.
